921 Inhibition of IARS alleviates skin inflammation in psoriasis through JAK2/STAT3 pathway

The aminoacyl-tRNA synthetases (ARSs) are a family of essential enzymes that participate the protein synthesis through ligate amino acids to their corresponding tRNAs. Although we have previously revealed mupirocin blocks Imiquimod (IMQ) -induced psoriasis-like skin lesion by inhibiting epidermal isoleucyl-tRNA synthetase (IARS), mechanisms IARS in psoriatic local inflammation remained unelucidated. Here, investigated was specifically highly expressed keratinocytes compared with immune cells. Importantly, found subcutaneous injection antibody alleviated epidermis hyperplasia and inflammatory infiltration imiquimod-induced mouse model. In vitro, silencing primary reduced expression related pro-inflammation cytokines, chemokines, antimicrobial peptides, such as IL-1β IL-6, LL37, CCL5, S100A7. Furthermore, observed inhibited cell proliferation promoted apoptosis via JAK2/STAT3 signaling pathway. Taken together, this study identifies critical role pathogenesis psoriasis proposes potential therapeutic target for psoriasis.
